Standard Operating Procedures:

 

 Diaminobenzidine

 

#1 Process:

 

   Use of Diaminobenzidine (DAB) as a substrate in

   immunocytochemistry techniques.

 

#2 Hazardous Chemicals/Class of Hazardous Chemicals:

 

   Diaminobenzidine, benzidine, Class: Carcinogen.

 

#3 Personal Protective Equipment:

 

   a) latex gloves, lab coats, eye protection (goggles) are to be worn during all procedures involving DAB.

 

#4 Engineering/Ventilation Controls:

 

   All diluting and aliquoting of DAB solutions will be carried out inside of the fume hood.

 

#5 Special Handling Procedures and Storage Requirements:

 

   a) - DAB powder is stored in -20 oC freezer (25 gram vials).

      - Stock solutions (10 mg/ml) in 500 ul aliquots are stored at -20 oC

     

   b) - The weighing out of powdered DAB, mixing of stock solutions, and all aliquoting of stock will be performed solely by Dr. Masliah (or Anthony Adame). 

 

      - The lab will be cleared of all personnel during the weighing, mixing, and aliquoting of DAB stock.

 

#6 Spill and Accident Procedures:

 

  Currently under evaluation (March 2003)

 

#7 Waste Disposal:

 

Bag and tag as hazardous chemical waste. Call EH&S for disposal.

 

#8 Approval Required:

 

Lab personnel using DAB are required to obtain prior approved of the P.I prior to working with DAB.

    Fume Hood, weighing bench, East bench (MTF 332).
